RESUMO

Despite several decades of mass drug administration and elimination-related activities, human onchocerciasis still represents a major parasitic threat in endemic regions. Among the challenges encountered by the elimination program is the lack of a suitable diagnostic tool that is accurate and non-invasive. Currently used methods are either invasive or not suitable for monitoring large numbers of patients. Herein, we describe the identification and characterization of Onchocerca volvulus heat shock protein 70 (OvHSP70) as a novel diagnostic biomarker for human onchocerciasis, which can directly be detected in urine samples of infected patients. This nematode-specific antigen was identified through LC-MS after differential SDS-PAGE using urine-derived protein extracts from O. volvulus-infected patients in Cameroon. Polyclonal antibodies generated in rabbits after cloning and expression of OvHSP70 in Escherichia coli reliably differentiated between urine samples from infected- and uninfected patients in a hypoendemic area of human onchocerciasis. These results provide an excellent basis for further development of a non-invasive and scalable diagnostic assay for human onchocerciasis using urine samples. Such a urine-based diagnostic assay will be of major importance for the elimination program of human onchcerciasis in endemic countries.

RESUMO

The use of green methods for ruthenium oxide nanoparticles (RuONPs) synthesis is gaining attention due to their eco-friendliness, cost-effectiveness, and availability. However, reports on the green synthesis and characterization of RuONPs are limited compared to other metal nanoparticles. The green synthesis and characterization of RuONPs using water extracts of Gunnera perpensa leaves as a reducing agent is reported in this study. The RuONPs were characterized using X-ray diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), and Ultraviolet spectroscopy (UV-VIS). MTT assay was used to assess the cytotoxicity of the RuONPs against MCF7 and Vero cell lines. X-ray diffraction analysis results revealed the presence of crystalline and amorphous forms of RuONPs, while IR spectroscopy revealed the presence of functional groups associated with G. perpensa leaves. SEM showed that the RuONPs consisted predominantly of hexagonal and cuboid-like structures with a considerable degree of agglomeration being observed. The cell culture results indicated a low anticancer efficacy of RuONPs against MCF7 and Vero cell lines, suggesting that RuONPs may not be a good lead for anti-cancer drugs. This study highlights the potential of using green synthesis methods to produce RuONPs and their characterization, as well as their cytotoxicity against cancer cells.

RESUMO

The nanostructures on the wings of Idea malabarica (Moore, 1877) were analysed using scanning electron microscopy, energy dispersive X-ray spectroscopy, atomic force microscopy, Fourier transform-infrared spectroscopy, and reflectance measurements. The chemical and morphological analyses revealed the chitin-based intricate nanostructures. The influence of the nanostructures on the wetting characteristics of the wing was investigated using optical imaging. Applying the Maxwell-Garnet approximation to the porosities within the nanostructures, the refractive indices, which relate the reflectance response, were estimated. It was concluded that the colour seen on the wings of the Idea malabarica originate from the nanostructural configurations of the chitin-based structures and the embedded pigment.
